The one on the passenger side is ported vacuum (only has vacuum when you open the throttle) and is normally used for the vacuum advance on the distributor. The one on the drivers side is manifold vacuum and has vacuum all of the time. It can be used for vacuum accessories such as the modulator valve or the hot-air flap on the factory air cleaner(which you will need to keep the carb from icing up in the winter). If you do not need another manifold vacuum port, you can just plug it.
