Have you tried stepping on the brake a few times while moving the shifter back and forth? There is a solenoid that unlocks the shifter, controlled by the brake pedal switch, and it can stick, or the brake switch may not be working.
Check your brake lights.

Try checking fuse #6 which protects the shift interlock, speed control and DRL module if installed.
Experience this same condition once when pulling over to stop and take a cell phone call. Lucky for me I a fuse chart print out and quickly found the blown fuse-----replaced it and was back on my way.
It would be horrible having this happen and not able to figure it out--towing and diagnosis would probably cost $250-400---all for a $.50 fuse!
Hope this is the fix!

Battery disconnect fuse 6 removal
I first disconnected my battery. While disconnected my shifter was finally able to move. I then shifted into neutral with my emergency brake on reconnected my battery then started my 2001 e150. I then removed fuse 6 to make a few stops before I checked this out further

I'm passing thru ..excuse the ambiguity. There's a gizmo under the shifter cover 'protecting' the vehicle from 'incidents' involving the key n theft...? if this is disabled, I used a hammer, then these lock up situations decrease to a mild PITA.
Try not to stomp n pressure the brake pedal as too much pressure may caws a longer time delay while the system readjusts.

Same thing-- Easy fix for my E250 stuck in park
I have a 2001 E250 I had the same problem. I replaced blown fuse #11 and it fixed the problem. I had a trailer wire problem that made the fuse blow. I hope this post might help someone searching with the same problem I had! S
