I know that if you have a c6, you will probally have to get another one, because the c6 for the inline6 300(or atleast on mine) are narrower then other c6's. So it wont bolt up to the engine. and as far as getting parts, you can more then likely just go to the ford house and order parts. Shop around though, and look for bargains.
Yes you will have to get a bigger raditor, hoses, new ingnition system, etc. Springs will do just fine. The rear end you might want to upgrade to some better gears. If I read right(and i have yet to check mine) the inline 6 came with 2.73 gears. On my truck, f150 250 are the same for the most part, and you can buy aftermarket parts for the f250 and they will work on the f150.
I have been known to be wrong, so just dont go off taking my word for everything i just posted.

I have an 83 F100 2WD SWB that had a 300 w/AOD. I swapped it for a 460 and a C6. I kept the original 3.08 rear but switched rad, engine, mounts, dual exhaust w/headers, Holley fuel injection, fuel tank, drive shaft...... the 300-6, 289, 302, 351 windsor, and 351 cleveland all have the same bellhousing bolt pattern. the 351 morphodite, 400, 429, and 460 all have the same bellhousing bolt pattern. but if you already have a c6 and you just swap them out you shouldnt have to change the crossmember unless you scoot the tranny forwards or backwards and still then id say all you will need to do will put new holes in the frame. hope ive been of some help.

Hoses, Complete Exhaust System,
Accessory Brackets, Transmission to C6,
Shorten Drive Shaft 3". Longer Gear Selector Rod.
Kick Down Rod. etc.
