Advantage of limited slip: More street friendly...it does allow the inside tires to turn slower on turns when you are on the throttle.
Advantage of locker: Ultimate traction. On the street, if you corner hard you will chirp the inside tire. When you are on the throttle with a Detroit Locker it locks both axle shafts together, when you coast they unlock. On a long wheel base vehicle like a SD especially with an auto trans, it's not as big a deal as on say a stick shift jeep.
Of course you also have selectable lockers like ARBs, that are basically an open diff when they are turned off, but flip the switch in the cab and it's like a spool.......axle shafts are always locked together when they are on.
