No. Not required ,
Although not required, I run rotella 5w synthetic found at Walmart and I grab a filter while I am there motorcraft 1999 I believe the number is . Testing got me me to be able to run 10k miles between changes
Trans fluid I run regular atf found at napa . But other guys get creative with Trans fluid too. Probably better But not required .

How many quarts oil and tranny fluid, and Mark will the synthetic help these tranny at all , I know I blew up 3 of them in a previous super duty ! I remember reading always to park where when first warmed up you have to throw it in reverse first as this keeps fluid pumping thru the trouble areas versus always driving away in drive helps them last ? Also heard of using 6.0 cooler ?

Up to 17 quarts of trans fluid if you drain the pan and the converter. If you use my fluid exchange CLICK HERE you'll change even more.
I think it is good insurance. When I had a 4R100 that I used for towing I ran Mobil 1 Synthetic ATF.
That only works if you are waving a rubber chicken out of the passenger's window from when you start the engine until the trans reaches 150F. If you are not waving the rubber chicken then this reverse first stuff is just superstition.
Yes, I have. I've suggested it a few hundred times.
Replace the air to oil cooler with a 6.0L cooler. Keep the radiator cooler in the circuit, and if your radiator doesn't have a cooler, you need a new radiator.

Like Mark said above Merc V is the specified fluid for the 4r100. I personally have Wally World Dex/Merc in mine and haven't had an issue. Keep it cool and clean and your 4R100 will treat you well.
