Theoretically, if you had good headers with a tuned exhaust, 2wd (strictly for less weight), timing bumped 13-15*, maf, a gear vendor (to get double od), tonneau cover to cut the wind, delete smog crap, run electric fans, and you have larger tires but find a way to lower the rig a bit to cut wind resistance, you could hit 15mpg.
But that's a theory. My best was 13mpg. 55mph cruising for 250 miles on a 19 gallon tank.

former farm truck, 96 f250 2wd CC 7.5efi ZF 3.55s 235/85/16 10ply allseasons. got it with 260,000km run beautiful, pass etest flying colours consistent 6.5-7mpg, never was driven over 60mph. we were afraid to put a trailer behind it!!!
we only used it for a year, its still waiting for its diesel conversion.

I think it can definitely be tuned for more power or torque. but I don't think without changing out the cam, and doing a port and polish on the intake manifold and Head, could you really get a significant change for mpg, at least not by just the computer.

This is what I had...down from Spokane to Moab and back and got 11.9, sitting right around 60mph the whole way (pulling about 6k lbs). A road trip empty from Spokane to Seattle at the same speed empty got me the same mpg (11.9). I put premium in for a trip once and I got 13.5 empty, but that was only one trip.

92 SuperCab LB 4x4 460. With 33" tires (speedometer corrected w PSOM), E4OD and 3.55 gears, I CAN get 14 mpg. On level highway, 60-65 mph and driving like I have an egg on the throttle. Still gets 10 towing 5-7000 pounds IF you take it easy. 266K on stock engine also. I tried timing bump to 14 then 12 and went back to 10 so I can run 87 octane fuel.

Well right when I got my F250 I got averaged 11-12 and best of 14. After I did my L&L headers, smog pump delete, exhaust and synthetic oil in the engine and trans, I hit 16 once driving 55-65 and got 12.5 pulling a mustang. The average was like 12-13. It seem no matter I'm hot rodding it or pulling not so big a load it will get 10. When I pulled my F350 CC 7.3 home with a 30 ft goose neck trailer it got 8. The pickup is all stock besides the headers has no AC or cruise and has 3.55s. It will cruise down the at 2k RPMs at 70MPH
